In the anime, Haori appears in Episode 461 "Hagoromo and Hamura". A relatively beautiful woman, Haori's significant appearence is demonstrated when she is seen walking on the path to the Divine Tree (also known as the God Tree) by Hagoromo and Hamura. Hagoromo and Hamura, who are shocked to see her, then question her on what she is doing. Haori responds by informing the two brothers that she has received notice to serve the Divine Tree, which is a ceremony instructed by the brothers' mother Kaguya, that began eons ago. Hagoromo, who denies that she goes to the tree, is given a red magatama amulet by Haori, which resembles the other six black magatamas found on Hagoromo's robe. Futhermore, Haori tells Hagoromo and Hamura that although she has only known them for a brief period of time, she is glad she has met them, and then continues on her path to the Divine Tree, despite Hagoromo calling out "Haori!"

Haori was a resident of the Land of Ancestors during the reign of Kaguya Ōtsutsuki.
After Hagoromo daringly decides to investiage the Divine Tree's truth (as told by Gamamaru) with his brother Hamura, they both discover through Hamura's Byakugan that the Divine Tree's truth is a large scale burial ground, with the deceased wrapped similar to the wrapping of a mummy. Hamura, who pinpoints Haori's body, instructs Hagoromo to break the wrapping around Haori's body. Hagoromo, in denial, repeatedly asks Haori to "wake up," but Hamura frankly tells Hagoromo that it's no use, and that Haori is dead. Hagoromo, who lets out his misery through a large yell, awakens the Sharigan in both of his eyes. Unique to Hagoromo, his two sharigans awakened to their full form, posessing the three tomoe in both eyes.

Hagoromo's response to Haori's death indicates how much he truly loved her, which led to the awakening of his Sharigans.
